  1. Indications and Usage Eltrombopag olamine tablets are indicated for the treatment of chronic immune thrombocytopenia (ITP) in adults and pediatric patients 6 years of age and older who have had an insufficient response to previous treatment with corticosteroids, immunoglobulins, or other therapies.
  2. Dosage and Administration Recommended Dosage: The recommended starting dose for adults and pediatric patients 12 years of age and older is 50 mg once daily. For pediatric patients 6 to 11 years of age, the recommended starting dose is 25 mg once daily. Administration: The tablet should be swallowed whole with water. It should be taken at least 2 hours before or 4 hours after taking antacids or containing polyvalent cations, or consuming dairy products. Dose Modification: The dose should be adjusted based on the patient’s platelet count response. The maximum recommended dose is 75 mg once daily for adults and 50 mg once daily for children 6 to 11 years of age.
  3. Mechanism of Action Eltrombopag is an orally available, small-molecule antagonist of the thrombopoietin (TPO) receptor. It stimulates megakaryocyte proliferation and differentiation, leading to increased platelet production.
  4. Safety and Warnings Thrombotic/Thromboembolic Complications: Serious thrombotic or thromboembolic complications, including portal vein thrombosis, have been reported. Hepatotoxicity: Elevations in liver enzymes and bilirubin have been observed. Liver function tests should be monitored regularly. Risk of Neoplasms: Long-term stimulation of the TPO receptor may increase the risk of neoplasia. Drug Interactions Polyvalent Cations: Antacids, laxatives, and supplements containing calcium, iron, magnesium, selenium, or zinc can significantly decrease absorption and should be separated by at least 2 hours before or 4 hours after administration. CYP2C8 Inhibitors: Concomitant use with gemfibrozil increases eltrombopag exposure and should be avoided.
