Motilium Domperidone Tablets

1. Indications and Usage
Indigestion Symptoms: Indicated for dyspeptic symptoms such as abdominal fullness, abdominal pain, belching, excessive intestinal gas, nausea, and vomiting.
Nausea and Vomiting: Indicated for nausea and vomiting caused by various factors including functional, organic, infectious, dietary causes, or radiation therapy and chemotherapy-induced nausea and vomiting.
2. Dosage and Administration
Recommended Dose: For adults, the typical dosage is 1 tablet (10 mg) once, 3 times daily.
Timing: Should be taken 15 to 30 minutes before meals.
Duration: Treatment duration should generally not exceed 1 week.
3. Mechanism of Action
D2 Antagonism: Domperidone selectively antagonizes peripheral dopamine D2 receptors.
Prokinetic Effect: It increases gastrointestinal motility and tone, promotes coordinated pyloric contraction, enhances esophageal peristalsis, and inhibits nausea and vomiting.
Blood-Brain Barrier: It has limited ability to cross the blood-brain barrier, resulting in fewer central nervous system adverse reactions compared to other antiemetics.
4. Safety and Warnings
Cardiac Risks: There is a risk of serious ventricular arrhythmias and sudden cardiac death, particularly in patients with heart disease, electrolyte imbalances, or doses exceeding 30 mg/day.
Hyperprolactinemia: May cause elevated serum prolactin levels, leading to galactorrhea, gynecomastia, and menstrual irregularities.
Contraindications: Contraindicated in patients with mechanical gastrointestinal obstruction, gastrointestinal hemorrhage, prolactin-secreting pituitary tumors, moderate to severe hepatic impairment, and known QTc prolongation.
5. Adverse Reactions and Clinical Research
Adverse Reactions: Common adverse reactions include headache, dry mouth, insomnia, abdominal cramps, diarrhea, and flatulence.
Clinical Research: Clinical trials have demonstrated its efficacy in accelerating gastric emptying and providing symptomatic relief for functional dyspepsia and acute nausea/vomiting.
6. Drug Interactions
QTc-Prolonging Agents: Concomitant use with strong CYP3A4 inhibitors (e.g., oral ketoconazole, erythromycin, clarithromycin) or other drugs that prolong the QTc interval is contraindicated.
Anticholinergics: May antagonize the prokinetic effects of domperidone.
Acid-Reducing Agents: Antacids or drugs that reduce gastric acidity may decrease the bioavailability of domperidone.
Levodopa: Domperidone can reduce peripheral side effects of levodopa without interfering with its central therapeutic effects.

Composition: Each tablet contains 10 mg of domperidone.
Excipients: Includes microcrystalline cellulose, starch, povidone, silica, magnesium stearate, and film-coating agents.
Appearance: White tablets.
