The study is a randomized, multicenter, double-blind, placebo-controlled clinical trial (n=420) designed to evaluate the efficacy and safety of BHV-1400 (500 mg subcutaneous injection once every two weeks) in patients with IgA nephropathy (IgAN). The primary endpoint is the change from baseline in urine protein-to-creatinine ratio (UPCR) at Month 52.<\/p>\n\n\n\n
BHV-1400 is a targeted removal of aberrant protein (TRAP) degrader developed by Biohaven based on its Molecular Degradation of Extracellular Proteins (MoDE) technology platform. According to Biohaven, existing IgAN therapies have varying degrees of impact on normal immunoglobulin levels, whereas BHV-1400 can precisely reduce pathogenic antibody levels in IgAN while maintaining normal immunoglobulin levels within the normal range.<\/p>\n\n\n\n
First-in-human Phase I data disclosed in January 2025 showed that BHV-1400 (125 mg, the lowest dose) reduced pathogenic galactose-deficient IgA1 (Gd-IgA1) antibody levels in IgAN patients by 60% within 4 hours of a single dose, up to 70% within 8 hours, with the effect lasting for several days. At the same time, BHV-1400 maintained immunoglobulin levels including IgA within the normal range.<\/p>\n\n\n\n
Data from an expanded cohort updated in May of this year showed that after one month of treatment with BHV-1400 (500 mg subcutaneous injection once every two weeks), Gd-IgA1 antibody levels in IgAN patients were reduced by 70%.<\/p>\n\n\n\n
IgA nephropathy is a progressive, autoimmune chronic kidney disease with onset typically between 20 and 40 years of age.
