1. Indications and Usage
\nNon-Small Cell Lung Cancer (NSCLC):
\nNon-Squamous NSCLC: Indicated in combination with pemetrexed and carboplatin for the first-line treatment of patients with metastatic non-squamous NSCLC who are negative for EGFR gene mutations and ALK gene rearrangements.
\nSquamous NSCLC: Indicated in combination with paclitaxel and carboplatin for the first-line treatment of patients with metastatic squamous NSCLC.
\nStage III NSCLC: Indicated for the treatment of patients with unresectable Stage III NSCLC who have not progressed after concurrent or sequential chemoradiotherapy.
\n2. Dosage and Administration
\nRoute of Administration:
\nIntravenous Infusion ONLY. Do not administer as an IV push or bolus.
\nRecommended Dosage:
\nDose: 1200 mg administered intravenously every 3 weeks.
\nDuration: Infuse over 60 minutes or longer. Continue until disease progression or unacceptable toxicity.
\nPreparation:
\nDilute the required volume into a sterile infusion bag containing 0.9% Sodium Chloride Injection (250 mL).
\n3. Mechanism of Action
\nPD-L1 Blockade:
\nSugemalimab is a human immunoglobulin G4 (IgG4) monoclonal antibody that binds directly to PD-L1.
\nImmune Activation:
\nIt inhibits the interaction between PD-L1 and its receptors (PD-1 and B7.1) on T cells and antigen-presenting cells, thereby blocking immunosuppressive reactions, including anti-tumor immune responses.
\n4. Safety and Warnings
\nImmune-Mediated Adverse Reactions:
\nSugemalimab can cause severe and life-threatening immune-mediated adverse reactions in any organ system, including pneumonitis, hepatitis, nephritis, and endocrinopathies (e.g., hypothyroidism). Monitor patients closely.
\nEmbryo-Fetal Toxicity:
\nMay cause fetal harm. Women of reproductive potential should use effective contraception during treatment and for at least 6 months after the last dose.
\n5.Adverse Reactions
\nMost Common:
\nFatigue, rash, hyperglycemia, musculoskeletal pain, abdominal pain, hypothyroidism, and elevated transaminases (ALT\/AST).
\n6. Drug Interactions
\nImmunosuppressants:
\nAvoid the use of systemic corticosteroids and other immunosuppressive drugs prior to initiating Sugemalimab, as they may interfere with its pharmacodynamic activity.
\nMetabolic Interactions:
\nAs a monoclonal antibody, Sugemalimab is not metabolized by CYP450 enzymes.
