Jiaxun Docusate Sodium and Dantron Capsules

1. Indications and Usage
Constipation: Indicated for the relief of constipation in pregnant women, patients with hemorrhoids, the elderly and weak, cardiovascular patients, and patients post-anal or gynecological surgery who need to avoid straining during defecation.
2. Dosage and Administration
Adults: The usual dosage is 1 to 2 capsules taken orally once daily.
Titration: After 3 days, the dosage should be reduced to 1 capsule daily as a maintenance dose based on patient response.
Duration: Long-term use is not recommended; continuous treatment should generally not exceed 1 week.
3. Mechanism of Action
Fecal Softening: Docusate sodium is a non-absorbable anionic surfactant that lowers the surface tension of feces, allowing water and fats to penetrate and soften the stool.
Stimulation of Peristalsis: Dantron (an anthraquinone glycoside) acts specifically on the large intestine, stimulating the intestinal nerve plexus to increase peristalsis and reduce water and sodium absorption in the bowel.
4. Safety and Warnings
Contraindications: Contraindicated in women during menstruation or pregnancy. Do not use in cases of abdominal pain or known hypersensitivity.
Toxicity and Carcinogenicity: Animal studies have linked dantron to intestinal adenocarcinoma and liver tumors. While no human cancer cases have been reported, the theoretical risk cannot be excluded.
Discoloration: The metabolites of dantron are excreted in urine and feces, which may cause orange discoloration and potentially perineal erythema in patients with incontinence.
Electrolyte Imbalance: Frequent or excessive use can lead to mild diarrhea, water and electrolyte disturbances, colonic inertia, and melanosis coli.
5. Adverse Reactions and Clinical Research
Common Adverse Reactions: Occasional mild abdominal cramping or diarrhea may occur.
Serious Effects: Long-term or high-dose use may cause skin rashes, skin irritation, skin discoloration, peeling of the perianal skin, leukopenia with liver damage, gray-blue skin color, and orange vaginal discharge.
6. Drug Interactions
Mineral Oil: Concomitant use with mineral oil is contraindicated, as it promotes the absorption of mineral oil into the intestinal mucosa, potentially leading to adverse reactions and toxicity.
7. Pharmaceutical Information
Active Ingredients: Each capsule contains Docusate Sodium 60 mg and Dantron 25 mg.
Appearance: Capsules containing yellow granules.
Storage: Store in a light-resistant, tightly closed container, protected from moisture.
Expiry: 24 months.
