1. Indications and Usage
Community-Acquired Pneumonia: Indicated for the treatment of mild to moderate community-acquired pneumonia (CAP) caused by sensitive bacteria in adult patients (≥18 years old).
2. Dosage and Administration
Route of Administration: Oral administration.
Recommended Dosage: 0.5 g once daily.
Administration Details: The treatment duration is typically 7 to 10 days. It is recommended to administer the capsule orally on an empty stomach, at least 1 hour before or 2 hours after meals.
Renal/Hepatic Impairment: No dose adjustment is required for patients with creatinine clearance ≥50 mL/min or mild to moderate hepatic impairment.
3. Mechanism of Action
DNA Gyrase Inhibition: Nemonoxacin inhibits bacterial DNA gyrase (topoisomerase II).
Topoisomerase IV Inhibition: It also inhibits topoisomerase IV.
Bactericidal Effect: The inhibition of these two essential enzymes prevents DNA replication and transcription, exerting a bactericidal effect against pathogens.
4. Safety and Warnings
QT Prolongation: May prolong the QT interval; caution is advised in patients with a history of QT prolongation or those taking antiarrhythmics.
Tendon Disorders: Associated with tendonitis and tendon rupture; discontinue immediately at the first sign of tendon pain or inflammation.
CNS Effects: May cause dizziness, confusion, or seizures. Use with caution in patients with a history of epilepsy.
Photosensitivity: Photosensitivity reactions have been reported; patients should avoid excessive exposure to sunlight or UV light.

Most Common Adverse Reactions: Gastrointestinal disturbances, skin reactions (rash, pruritus), and CNS effects (dizziness, headache).

6. Drug Interactions
Chelating Agents: Antacids, iron, calcium, magnesium, or zinc supplements may reduce absorption and should be administered at least 2 hours after nemonoxacin (except calcium supplements, which can be co-administered).
NSAIDs: Concomitant use with non-steroidal anti-inflammatory drugs (NSAIDs) may increase the risk of CNS excitation and seizures.
Theophylline: Potential interaction leading to increased serum theophylline levels and associated adverse effects.
