Recommended Dosage: The recommended dosage is 100 mg administered by subcutaneous injection at week 0 and week 4, followed by every 8 weeks thereafter.
Administration: Avoid injecting into areas where psoriasis is present. Patients may be instructed to self-inject after proper training.
Monitoring: Patients who do not respond adequately by week 16 should be reassessed, and discontinuation should be considered.
3. Mechanism of Action
Guselkumab is a human monoclonal antibody that selectively targets the p19 subunit of interleukin-23 (IL-23). By binding to IL-23, it inhibits the differentiation and proliferation of T cells, thereby reducing the inflammatory response associated with plaque psoriasis.
4. Safety and Warnings
Infections: Guselkumab may increase the risk of infections. Patients with important clinically significant active infections, including active tuberculosis, should not initiate therapy.
Hypersensitivity: Serious hypersensitivity reactions have been reported.
Vaccinations: Live or live-attenuated vaccines should be avoided during treatment with guselkumab.
