1. Indications and Usage
Peptic Ulcer Disease: Treatment of gastric and duodenal ulcers.
2. Dosage and Administration
Adults: Oral administration.
Dosage: 1.5 g (three 0.5g tablets) per dose, twice daily.
3. Mechanism of Action
Mucosal Protection: Forms a viscous suspension in gastric juice, which physically protects the ulcerated area.
Enzyme Inhibition: Reduces pepsin activity, thereby decreasing proteolytic degradation of the gastric mucosa.
Pharmacokinetics: Has very low oral bioavailability and acts locally within the gastrointestinal tract.
4. Safety and Warnings
Concomitant Medication: Should not be taken simultaneously with other medications, as it may interfere with their absorption.
Special Populations: Safety and efficacy in pregnant and lactating women have not been fully established.
5. Adverse Reactions and Clinical Research
Gastrointestinal Effects: Occasional nausea, vomiting, abdominal pain, diarrhea, gastric fullness, acid reflux, heartburn, and hiccups.
Other Effects: Constipation and skin rashes may occur rarely.
6. Drug Interactions
Proton Pump Inhibitors/Antacids: Take at least 30 minutes after Dosmalfate.
Fluoroquinolone Antibiotics: Take 2 to 6 hours before or 4 hours after Dosmalfate to prevent insoluble compound formation.
Antifungals (e.g., Ketoconazole): Take at least 2 hours before Dosmalfate.
Cardiac Drugs (e.g., Digoxin, Quinidine): Take at least 2 to 4 hours before Dosmalfate.
Tetracyclines: Take 2 to 3 hours before Dosmalfate.
Other Drugs: May reduce the pharmacological effects of oral statins (e.g., clofibrate), anticoagulants (e.g., warfarin), and oral hypoglycemics (e.g., tolbutamide).
