- Indications and Usage Influenza A and B: Indicated for the treatment of acute, uncomplicated influenza A and B infection in patients 5 years of age and older. Treatment Window: Efficacy is maximized when administered within 48 hours of the onset of influenza symptoms.
- Dosage and Administration Adults and Adolescents (≥12 years): 200 mg taken orally once daily for 5 days. Pediatric Patients (5 to 11 years): 120 mg taken orally once daily for 5 days. Administration Details: The tablet can be taken with or without food. Renal/Hepatic Impairment: No dosage adjustment is required for mild to moderate hepatic impairment. For patients with severe renal impairment (eGFR <30 mL/min/1.73 m²), a dosage reduction to 100 mg once daily is recommended.
- Mechanism of Action PA Inhibition: Suraxavir marboxil is a prodrug that is rapidly hydrolyzed in vivo to its active metabolite, suraxavir. Viral Replication Blockade: Suraxavir selectively and competitively inhibits the cap-snatching endonuclease activity of the influenza virus PA protein. This blocks viral mRNA transcription and subsequent viral genome replication.
- Safety and Warnings Neuropsychiatric Events: Influenza infections themselves can cause confusion or hallucinations. Patients should be monitored for abnormal behavior, particularly in children and adolescents. Hypersensitivity: Serious skin reactions (e.g., Stevens-Johnson syndrome) have been reported with antiviral agents. Vaccine Efficacy: The drug may reduce the effectiveness of live attenuated influenza vaccines.
- Adverse Reactions and Clinical Research Most Common Adverse Reactions: Nausea, vomiting, diarrhea, and headache. Clinical Research: Clinical trials have demonstrated that suraxavir marboxil significantly shortens the duration of influenza symptoms and reduces viral shedding compared to placebo.
- Drug Interactions P-gp Inhibitors: Strong P-glycoprotein (P-gp) inhibitors (e.g., ketoconazole, cyclosporine) may increase suraxavir exposure. P-gp Inducers: Strong P-gp inducers (e.g., rifampin, phenytoin) may decrease suraxavir exposure. Live Vaccines: Concomitant use with live attenuated influenza vaccine is not recommended.
- Pharmaceutical Information Chemical Composition: Active ingredient: Suraxavir marboxil. Appearance: Film-coated tablets. Packaging: Supplied in blister packs. Storage: Store at controlled room temperature (20°C to 25°C). Protect from moisture.
