1. Indications and Usage
Gastrointestinal Disorders: Treatment of functional dyspepsia accompanied by gastrointestinal symptoms such as gastric burning, belching, nausea, vomiting, early satiety, and upper abdominal distension.
2. Dosage and Administration
Adults: The usual dosage is 1 capsule (5 mg) taken 3 times daily.
Administration: Capsules should be swallowed with water before meals.
Duration: If gastrointestinal symptoms do not improve after approximately 2 weeks of treatment, the medication should be discontinued.
3. Mechanism of Action
5-HT4 Receptor Agonism: Mosapride citrate acts as a selective agonist at serotonin type 4 (5-HT4) receptors in the myenteric plexus of the gastrointestinal tract.
Acetylcholine Release: Stimulation of these receptors promotes the release of acetylcholine, thereby enhancing gastrointestinal motility and accelerating gastric emptying without affecting gastric acid secretion.
4. Safety and Warnings
Contraindications: Contraindicated in patients with gastrointestinal hemorrhage, mechanical obstruction, or perforation.
Special Populations: Safety and efficacy in pregnant or breastfeeding women and children have not been fully established; use should be avoided unless necessary.
Monitoring: Elderly patients should be monitored closely, and dosage should be reduced if adverse reactions occur.
5. Adverse Reactions and Clinical Research
Common Adverse Reactions: Diarrhea, abdominal pain, dry mouth, rash, and general fatigue or dizziness may occur occasionally.
Other Effects: Rarely, abnormalities in liver function tests (elevated AST, ALT, ALP, GGT), hypertriglyceridemia, eosinophilia, palpitations, or ECG changes may be observed.
6. Drug Interactions
Anticholinergics: Concomitant use with anticholinergic drugs (e.g., atropine) may reduce the prokinetic effects of mosapride. A time interval should be maintained if both are used.
Other Medications: Caution is advised when used with other drugs that may affect gastrointestinal motility or cardiac conduction.
