Alecensa® is the brand name developed by Roche. It is a highly selective anaplastic lymphoma kinase (ALK) tyrosine kinase inhibitor. It targets ALK fusion proteins to block downstream oncogenic signalling, suppressing tumour cell proliferation and inducing apoptosis.
Main Indications
For the treatment of patients with anaplastic lymphoma kinase (ALK)-positive locally advanced or metastatic non-small cell lung cancer (NSCLC).
Clinical Research
Global ALEX trial demonstrated that alectinib significantly prolonged progression-free survival compared with crizotinib in treatment-naive ALK-positive advanced NSCLC. It exhibits strong intracranial anti-tumour activity and effective control of brain metastases. Long-term follow-up showed sustained survival benefit.
Common Side Effects
Systemic reactions: fatigue, peripheral oedema;
Common mild adverse reactions: constipation, myalgia, elevated liver enzymes. Rare serious adverse events: interstitial lung disease, bradycardia.
