On June 15, the NMPA official website showed that Bristol Myers Squibb’s TYK2 inhibitor Deucravacitinib has been approved for a new indication in China for the treatment of psoriatic arthritis .
Deucravacitinib is a tyrosine kinase 2 (TYK2) inhibitor that selectively targets TYK2 to inhibit signaling of IL-23, IL-12, and IFN — cytokines that are key mediators involved in the pathogenesis of multiple immune-mediated diseases. Deucravacitinib achieves high selectivity by binding to the regulatory domain of TYK2, enabling allosteric inhibition of TYK2 and its downstream functions. Within the physiological concentration range, the product selectively inhibits TYK2 without inhibiting JAK1, JAK2, or JAK3 at therapeutic doses.
According to the Insight database, Deucravacitinib was first approved in the United States in September 2022 under the brand name Sotyktu for adult plaque psoriasis, becoming the world’s first approved TYK2 allosteric inhibitor. Subsequently, the drug has been approved in Japan, Canada, Australia, and the European Union.
