3. Mechanism of Action
Potassium-Competitive Acid Blockade: Tegoprazan reversibly inhibits the gastric H⁺/K⁺-ATPase (proton pump) in a potassium-competitive manner.
Rapid Onset: Unlike traditional proton pump inhibitors, it does not require acid-dependent activation, providing rapid and sustained acid suppression from the first dose.
4. Safety and Warnings
Hepatotoxicity: Liver function abnormalities, including elevated liver enzymes, have been reported. Liver function should be monitored regularly.
Hypersensitivity: Contraindicated in patients with known hypersensitivity to tegoprazan or benzimidazoles.
Masking Malignancy: May mask symptoms of gastric malignancy; malignancy should be ruled out before initiating treatment in patients with dyspeptic symptoms.
Infection Risk: Suppression of gastric acidity may increase the risk of gastrointestinal infections.
5. Adverse Reactions and Clinical Research
Most Common Adverse Reactions: Increased blood gastrin, metal taste, diarrhea, nausea, and abdominal discomfort.
Clinical Research Highlights: Clinical trials have demonstrated that tegoprazan provides effective symptom relief and mucosal healing in acid-related diseases with a favorable safety profile.
6. Drug Interactions
pH-Dependent Drugs: Tegoprazan increases gastric pH, potentially reducing the bioavailability of drugs requiring an acidic environment for absorption (e.g., ketoconazole, itraconazole).
CYP3A4 Inhibitors: Strong CYP3A4 inhibitors (e.g., ketoconazole, ritonavir) may increase tegoprazan exposure.
Contraindicated Drugs: Concomitant use with atazanavir, nelfinavir, or rilpivirine is prohibited due to significant interactions.
