1. Indications and Usage
Contezolid Tablets are indicated for the treatment of acute bacterial skin and skin structure infections (ABSSSI) caused by susceptible Gram-positive microorganisms, including methicillin-resistant Staphylococcus aureus (MRSA),Streptococcus pyogenes,Streptococcus agalactiae,Streptococcus dysgalactiae,Streptococcus pneumoniae, and Streptococcus agalactiae.
2. Dosage and Administration
Standard Dosage: The recommended dosage is 800 mg (two tablets) administered orally every 12 hours.
Duration: The suggested treatment duration is 7 to 14 days, which can be adjusted by the treating physician based on the infection site, severity, and patient response.
Administration: The tablets should be taken with food or within 30 minutes after a meal to ensure optimal absorption.
Special Populations: No dosage adjustment is typically required for patients with renal or hepatic impairment.
3. Mechanism of Action
Contezolid is a synthetic oxazolidinone antibacterial agent. It works by inhibiting the formation of the bacterial 70S initiation complex, thereby preventing the synthesis of essential bacterial proteins and inhibiting bacterial growth.
4. Safety and Warnings
Hypersensitivity: Contraindicated in patients with known hypersensitivity to contezolid, other oxazolidinones, or any excipients.
Pancreatitis: Acute pancreatitis has been reported with oxazolidinones. Patients should be monitored for symptoms of severe abdominal pain.
Microbiological Confirmation: To reduce the development of drug-resistant bacteria, Contezolid should only be used for infections confirmed or highly suspected to be caused by susceptible organisms. It is not effective against Gram-negative infections.
5. Adverse Reactions and Clinical Research
Adverse Reactions: Common adverse reactions include nausea, diarrhea, vomiting, headache, and injection site reactions (if transitioning from IV to oral).
Clinical Research: In pivotal Phase III clinical trials, Contezolid demonstrated non-inferiority to linezolid in terms of clinical cure rates for ABSSSI, with a lower incidence of hematological toxicity (e.g., thrombocytopenia).
6. Drug Interactions
Serotonergic Drugs: As with other oxazolidinones, caution is advised when used with selective serotonin reuptake inhibitors (SSRIs) or other serotonergic drugs due to the potential risk of serotonin syndrome.
Metabolism: Contezolid is primarily metabolized by flavin monooxygenase 5 (FMO5). Clinically significant interactions with CYP450 inhibitors or inducers are not anticipated.
